Summary
Takrolimus is a metabolite of the fungus Streptomyces tsukubaensis. FK506 is a powerful immunomodulatory macrolide that acts like cyclosporin A, resulting in a decreased production of IL-2, IL-3, IL-4, TNF -alpha and GM-CSF. Topically administered tacrolimus is associated with mild local effects and unlike glucocorticoids it does not produce skin atrophy.
Tacrolimus is used successfully in the treatment of atopic dermatitis. In numerous trials, takrolimus oinment has shown to be an effective therapy in other immunologically mediated skin diseases including: contact dermatitis, psoriasis, pyoderma gangrenosum, erosive mucosal lichen planus, vitiligo. Nevertheless, further studies are required to evaluate the efficacy and the long-term safety of this novel agent.
