Propozycja procedury oceny przeciwpłytkowych właściwości preparatów polifenolowych pochodzenia roślinnego w badaniach in vitro**

Summary
Polyphenolic compounds have pleiotropic effects including the ability to reduce platelet aggregation. However, a commonly approved procedure allowing to detect the antiplatelet properties of these compounds has not been established yet. The aim of our study was to identify the best methods (laboratory procedures) to study the impact of polyphenolic compounds contained in plant extracts on ADP-induced platelet reactivity. The experiment consisted of two steps. At first, from among four plant extracts (Aronox®, Pycnogenol®, grape seeds, raspberry seeds) we chose the extract having the strongest inhibitory effect on platelet aggregation. We decide to use the optical aggregometry as a scrreening method, since it is regarded as the gold standard in studies of platelets’ reactivity. In next step, we selected the most effective method of detecting in vitro antiplatelet properties of polyphenolic extracts. Totally, we compared: light transmission aggregometry (LTA), impedance agreggometry (MEA), platelet adhesion (Impact-R), expression of selected surface membrane platelet receptors (flow cytometry). In the first step of our experiment, the best inhibitor of platelet aggregation was grape seed extract. In the second step, we found that LTA was the best technique since it allowed to detect a significant inhibition of platelets under by grape seed extract (inhibition score 6). Thus, LTA defended its gold standard position. Our results suggested also that impedance aggregometry (inhibition score 4) was also useful in detection of antiplatelet effects of grape seed extract, whereas Impact-R and flow cytometry were not suitable methods in this case (inhibition score 1 and 0, respectively).
