Summary
Nimesulide is a preferential inhibitor of cyclooxygenase-2 (COX-2) with strong anti-inflammatory, analgesic and antipyretic activities. However, it appears to exert its effects also through some additional unique mechanisms. There are evidences that nimesulide poses chondroprotective activities. Although, its adverse events are similar to those of other non-steroidal anti-inflammatory drugs (NSAIDs), they were shown to be milder in intensity and less frequent. Nimesulide is well-tolerated and effective drug also in the geriatric population. This review describes the pharmacokinetics, mechanisms of action, safety and the applications of nimesulide in the treatment of rheumatic diseases.
